目的:探讨原发性大肠癌中CD44v mRNA表达与癌细胞DNA含量之间的关系及其临床意义.方法:应用RT-PCR法检测72例大肠癌中CD44v mRNA的表达情况,并根据CD44v mRNA的表达情况(阴性及微弱表达17例为Ⅰ组,中高度以上表达者55例为Ⅱ组)检测其癌细胞DNA干系水平.结果:72例受检标本中,CD44v mRNA中度以上表达55例(76.39%),其中有淋巴结转移者为93.62%(44/47),无淋巴结转移者为44.00%(11/25).在DNA干系水平方面,Ⅰ组病例二倍体明显高于Ⅱ组病例,异倍体则明显低于Ⅱ组病例(P<0.01).结论:CD44v mRNA的高表达与大肠癌的侵袭转移关系密切,CD44v mRNA表达与大肠癌细胞DNA干系水平的变化一致,再次证实了CD44v mRNA表达在大肠癌侵袭转移中的重要性
Objective: To investigate the relationship between the expression of CD44v mRNA and DNA content in primary colorectal cancer and its clinical significance. Methods: RT-PCR was used to detect the expression of CD44v mRNA in 72 cases of colorectal cancer, and according to the expression of CD44v mRNA. The expression status (negative and faint expression of 17 cases in group I and 55 cases of moderate or high expression in group II) was used to detect the level of DNA of the cancer cells. Results: Of the 72 specimens examined, 55 cases of moderate or high expression of CD44v mRNA ( 76.39%) of them had 93.62% (44/47) with lymph node metastases and 44.00% (11/25) with no lymph node metastases. In terms of DNA stemline level, diploids in group I were significantly higher than those in group II. The aneuploidy was significantly lower than that in the group II (P<0.01). Conclusion: The high expression of CD44v mRNA is closely related to the invasion and metastasis of colorectal cancer.
