目的 :了解头颈部鳞癌及其颈淋巴结转移癌端粒酶的表达情况 ,探讨端粒酶活性定量分析在头颈部鳞癌诊断中的价值。方法 :采用端粒重复序列液体闪烁计数法检测端粒酶活性。共检测取自 2 5例头颈部鳞癌患者的组织样本 5 5份 ,其中 7例患者同时取有原发癌及其颈淋巴结转移癌两份样本 ,以 2 3份正常组织作为对照。结果 :132份原发鳞癌组织中端粒酶活性 (cpm值 )在 10 0 0以上的 2 8份 ,除 2份外 ,均明显高于正常组织 ;2 3份正常组织的端粒酶活性均在 10 0 0以下 ,除 1份外均低于所有鳞癌组织 (P <0 .0 5 )。 2 7份颈淋巴结转移癌与其原发癌组织同样具有较高的端粒酶活性 (P >0 .0 5 )。结论 :1液体闪烁定量分析方法检测头颈部鳞癌端粒酶活性敏感性较高 ,对头颈部恶性肿瘤的辅助诊断具有一定价值。 2转移癌细胞同样具有较高的端粒酶活性。
Objective: To investigate the expression of telomerase in head and neck squamous cell carcinoma and cervical lymph node metastasis, and to explore the value of quantitative telomerase activity in the diagnosis of squamous cell carcinoma of the head and neck. Methods: Telomerase activity was detected by liquid scintillation counting of telomeric repeat sequences. A total of 55 tissue samples from 25 patients with squamous cell carcinoma of head and neck squamous cell carcinoma were screened. Seven of them had two samples of primary carcinoma and cervical lymph node metastasis at the same time, and 23 normal tissues were taken as control. Results: The telomerase activity (cpm value) in 132 primary squamous cell carcinomas was significantly higher than that in normal tissues except for 2 out of 28 samples with the cpm value above 100 000. The telomerase activity in 23 normal tissues All below 100, with the exception of 1, which was lower than that of all squamous cell carcinomas (P <0.05). Twenty-seven cervical lymph node metastases also had higher telomerase activity than their primary cancers (P> 0.05). Conclusion: 1 liquid scintillation quantitative analysis of head and neck squamous cell carcinoma of the telomerase activity of high sensitivity, head and neck cancer diagnosis of malignant tumors have a value. 2 metastatic cancer cells also have higher telomerase activity.
