目的观察中药雄黄灌胃(ig)给药对大鼠生育力与早期胚胎发育的毒性影响。方法 SD雌雄大鼠分为阴性对照组,雄黄低、中、高剂量组4组,雄性大鼠连续ig给药6周后与连续给药2周的雌鼠交配,每天ig给予批号为H2007052401雄黄250、125和50 mg.kg-13个剂量。雄性给药至交配全部结束,雌性给药至妊娠第6天。雄鼠交配后处死,雌鼠妊娠第15天处死,计算黄体数、着床数、活胎数等,并与阴性对照组比较。结果雄黄各剂量组动物的交配率、受孕率、活胎率、着床前后丢失率、黄体数、着床数以及生殖脏器方面与阴性对照组比较均未有显著差异;病理组织学方面:所有高剂量组及对照组动物的睾丸、附睾、卵巢未见明显异常。结论批号为H2007052401雄黄在本实验中对大鼠生育力和早期胚胎发育没有明显的毒性作用。
Objective To observe the toxic effects of administration of androgynoside (ig) on the fertility and early embryo development in rats. Methods SD male and female rats were divided into four groups: the control group, the low, middle and high dose of realgar; the male rats were mated with the female rats continuously administered for two weeks after the continuous ig administration for 6 weeks; the daily batch was H2007052401 realgar 250, 125 and 50 mg.kg-13 doses. Male dosing to the end of mating, female dosing to 6 days of pregnancy. The males were sacrificed after mating and the females were sacrificed on the 15th day of gestation. The number of luteal matings, the number of implants and the number of live births were calculated and compared with the negative control group. Results There was no significant difference in the mating rate, the pregnancy rate, the live fetus rate, the loss rate before and after implantation, the number of luteal and the number of implants and the reproductive organs in each dose of realgar. The histopathological aspects No significant abnormalities were observed in testis, epididymis and ovary in all high-dose and control animals. Conclusion lot number H2007052401 realgar in this experiment on rat fertility and early embryonic development has no significant toxic effects.
