目的:观察升清降浊方对皮下注射谷氨酸钠(MSG)大鼠骨骼肌的影响,以探索其改善胰岛素抵抗的作用机制。方法:新生大鼠皮下注射MSG并高脂诱导,制造胰岛素抵抗肥胖大鼠,选择48只MSG肥胖模型大鼠,雌雄各半。将大鼠随机分为6组:模型组,菲诺贝特100 mg.kg-1剂量组,罗格列酮5 mg.kg-1剂量组,升清降浊方6,3,1.5 g.kg-1剂量组;选正常鼠8只,雌雄各半做为正常对照组。给药50 d后,观察受试药物对MSG肥胖模型大鼠骨骼肌中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及糖元含量的影响。结果:升清降浊方对MSG肥胖模型大鼠骨骼肌糖脂代谢有明显改善作用,与模型组相比,给药组大鼠骨骼肌中TC,TG,LDL-C及糖元含量明显降低,升清降浊方3个剂量组具有极显著性差异(P<0.01);给药组骨骼肌中HDL-C含量明显升高,升清降浊方3 g.kg-1剂量组具有极显著性差异(P<0.01)。结论:升清降浊方能明显改善MSG大鼠骨骼肌组织中脂质和糖原含量。
Objective: To observe the effects of Shengqing Jiangzhuo prescription on the skeletal muscle of rats with subcutaneously injected glutamate (MSG) to explore its mechanism of improving insulin resistance. Methods: Newborn rats were injected subcutaneously with MSG and induced with high fat to produce insulin resistant obese rats. Forty-eight MSG obese rats were selected and divided into two groups, male and female. The rats were randomly divided into 6 groups: the model group, the fenofibrate 100 mg.kg-1 dose group, the rosiglitazone 5 mg.kg-1 dose group, Shengqing Jiangzhuo 6,3,1.5 g. kg-1 dose group; normal mice were selected 8, male and female as a normal control group. After administration for 50 days, the effects of test drugs on total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol -C) and glycogen content. Results: Shengqingjiangzhuo decoction could obviously improve the metabolism of skeletal muscle glycolipid in MSG model rats, and compared with model group, the content of TC, TG, LDL-C and glycogen in skeletal muscle of rats in treatment group were obviously decreased (P <0.01). The content of HDL-C in the skeletal muscle of the treated group was significantly increased, and the dose of Shengqing Jiangzhuo Decoction 3g.kg-1 had extremely significant difference Significant difference (P <0.01). Conclusion: Shengqing Jiangzhuo Fang can significantly improve the content of lipids and glycogen in skeletal muscle of MSG rats.
