目的研究香烟烟气对肺部炎性介质IL-1,IL-6,IL-8表达的影响。方法 (1)建立动物长期吸烟模型,用放射免疫法检测肺灌洗液(BALF)中IL-1,IL-6,IL-8的表达水平。(2)制备香烟烟气提取物(CSE)并用它染毒A549细胞,检测细胞上清液中IL-1,IL-6,IL-8的含量。结果动物吸烟各剂量组IL-1表达水平与对照组相比差异无统计学意义(P﹥0.05),低、中、高剂量组IL-6,IL-8水平显著升高,差异有统计学意义(P﹤0.05)。CSE染毒的A549细胞培养上清液中IL-1表达水平与对照组相比差异无统计学意义(P﹥0.05),20%、50%、100%CSE组IL-6的表达水平明显升高,IL-8表达水平下降,差异有统计学意义(P﹤0.05)。结论香烟对肺部炎性介质IL-6,IL-8的表达有影响。
Objective To study the effects of cigarette smoke on the expression of IL-1, IL-6 and IL-8 in pulmonary inflammatory mediators. Methods (1) The long-term smoking model was established in rats. The expression of IL-1, IL-6 and IL-8 in lung lavage fluid (BALF) was detected by radioimmunoassay. (2) Cigarette smoke extract (CSE) was prepared and used to infect A549 cells to detect the contents of IL-1, IL-6 and IL-8 in the supernatant. Results There was no significant difference in the expression of IL-1 between various smoking groups (P> 0.05), and the levels of IL-6 and IL-8 in low, medium and high dose groups were significantly increased Significance (P <0.05). The expression level of IL-1 in CSE-treated A549 cells was not significantly different from that in control group (P> 0.05). The expression level of IL-6 in CSE-treated A549 cells was significantly increased High, IL-8 expression decreased, the difference was statistically significant (P <0.05). Conclusion Cigarettes have an effect on the expression of IL-6 and IL-8 in the lung inflammatory mediators.
