目的 2008年对由于吉林省抚松县采集到的汉坦病毒阳性鼠肺组织进行病毒分离与基因鉴定。方法采集到鼠肺组织209份,使用直接免疫荧光法检测出汉坦病毒抗原阳性组织4份,对阳性组织采用Vero E6细胞进行病毒分离,对毒株进行血清学(酶联免疫吸附法和直接免疫荧光法)、分子生物学(聚合酶链式反应方法)检验,对基因序列使用MEGA 4.0分子生物学分析软件进行分析。结果实验检出汉坦病毒抗原阳性鼠肺4份,分离到汉城型病毒2株,命名为F1373和F1443。结论在抚松县采集的大林姬鼠标本中分离到2株病毒,经实验鉴定为汉坦病毒汉滩型。
Objective In 2008, virus isolation and gene identification of Hantavirus positive mice lungs collected from Fusong County of Jilin Province were performed. Methods A total of 209 rat lung tissues were collected. Four positive samples of Hantavirus antigen were detected by direct immunofluorescence. Vero E6 cells were used for virus isolation of positive tissues. The serological (enzyme-linked immunosorbent assay and direct Immunofluorescence), molecular biology (polymerase chain reaction method) test, the gene sequence using MEGA 4.0 molecular biology analysis software for analysis. Results Four Hantavirus positive mouse lungs were detected in the experiment. Two strains of Seoul virus were isolated and named as F1373 and F1443. Conclusions Two viruses were isolated from Apodemus agrarius collected in Fusong County and were identified as Hantaan hantavirus.
