目的:比较不同型号大孔吸附树脂对酸枣仁皂苷的纯化效果,并优选其纯化工艺。方法:以酸枣仁皂苷A为指标成分,HPLC-ELSD测定指标成分含量。通过静态吸附法和动态吸附法优选大孔树脂型号,采用正交试验和单因素试验考察其纯化工艺。结果:酸枣仁皂苷A在0.216~2.16μg与峰面积成良好线性关系,平均加样回收率为95.77%,RSD 0.71%。ADS-7型大孔吸附树脂能有效除去色素,纯化效果最好,其优化纯化工艺为柱高径比3:1,药液质量浓度1.0 g.mL-1,吸附速度1.0 BV.h-1,树脂生药吸附量0.4 g.g-1,洗脱溶媒70%乙醇,洗脱速度2 BV.h-1,洗脱溶媒用量4 BV。结论:ADS-7型大孔吸附树脂对酸枣仁皂苷A纯化效果较好,工艺稳定可行,可用于工业化生产。
Objective: To compare the purification effect of jujube jujube saponins by different types of macroporous adsorption resins, and to optimize its purification process. Methods: Jujuboside A was used as the index component, and the content of the index components was determined by HPLC-ELSD. Through the static adsorption method and the dynamic adsorption method, the macroporous resin model was optimized. The orthogonal experiment and single factor experiment were used to investigate the purification process. Results: Jujuboside A had a good linearity between 0.216 ~ 2.16μg and its peak area. The average recoveries were 95.77% and RSD 0.71%. ADS-7 macroporous resin can effectively remove the pigment, the best purification effect, the optimization of purification process for the column height to diameter ratio of 3: 1, liquid mass concentration of 1.0 g.mL-1, the adsorption rate of 1.0 BV.h-1 , Resin crude drug adsorption 0.4 gg-1, 70% ethanol elution solvent, the elution rate of 2 BV.h-1, the amount of solvent eluted 4 BV. Conclusion: The ADS-7 macroporous resin has good purification effect on jujuboside A. The technology is stable and feasible and can be used in industrial production.
