目的:探讨抑癌基因PTEN在肾盂癌中的表达及临床意义,及其与微血管密度(MVD)的关系。方法:采用免疫组织化学技术检测30例肾盂癌组织中PTEN及FⅧRag(MVD标记物)的表达,分析PTEN的表达与MVD及肾盂癌临床分期、病理分级的关系。结果:1PTEN蛋白在肾盂癌原发灶组织中较在良性病变肾脏组织中呈低表达(阳性表达率分别为86.67%和100%,P<0.05),且其表达率和肾盂癌临床分期及病理分级成负相关(P<0.01);2PTEN阳性表达率与FⅧRag阳性表达率呈负相关趋势(P<0.01)。结论:抑癌基因PTEN在肾盂癌组织中异常低表达,并和肾盂癌MVD及临床病理特征负相关;其可能是通过抑制血管生长来遏制肾盂癌的进展,有望为肾盂癌的诊治及预后提供新的指标及基因靶点。
Objective: To investigate the expression of tumor suppressor gene PTEN in renal pelvic cancer and its clinical significance and its relationship with microvessel density (MVD). Methods: The expressions of PTEN and FⅧRag (MVD markers) in 30 cases of renal pelvic carcinoma were detected by immunohistochemistry. The relationship between the expression of PTEN and the clinical stage and pathological grade of renal pelvis and carcinoma was analyzed. Results: The expression of PTEN protein in primary renal pelvis and cancerous tissues was lower than that in benign lesions (positive rates were 86.67% and 100%, respectively, P <0.05) (P <0.01). There was a negative correlation between the positive rate of 2PTEN and the positive rate of FⅧRag (P <0.01). CONCLUSION: PTEN, a tumor suppressor gene, is abnormally expressed in renal pelvis and cancerous tissues and is negatively correlated with MVD and clinicopathological features of renal pelvic carcinoma. It may be used to restrain the progress of renal pelvic cancer by inhibiting the growth of blood vessels and is expected to provide the diagnosis and treatment of renal pelvic cancer New indicators and gene targets.
