本文通过试验对原有分光光度法步骤进行优化,即Na Fe EDTA与硫氰酸钾反应生成红色配合物,最大吸收波长为λmax=490 nm。其中优化了Na Fe EDTA溶液的配置和使用乙醇代替甲醇两部分。Na Fe EDTA浓度在20.0~160.0μg/10 m L范围与吸光度呈良好的线性关系,相关系数为0.9997,在试样加标回收率为98.0%~106.0%,相对标准偏差(RSD)为1,.4%~3.9%。与卫生部推荐方法比较相差不大。
In this paper, the original step of spectrophotometry was optimized through experiments, that is, NaFe EDTA reacts with potassium thiocyanate to form a red complex. The maximum absorption wavelength is λmax = 490 nm. Which optimized the configuration of Na Fe EDTA solution and the use of ethanol instead of methanol in two parts. The concentration of NaFe EDTA in the range of 20.0 ~ 160.0 μg / 10 m L showed a good linear relationship with the absorbance, the correlation coefficient was 0.9997, the recoveries of spiked samples were 98.0% -106.0%, the relative standard deviation (RSD) was 1, .4% ~ 3.9%.
