利用肠道病毒(包括甲肝病毒)的3′末端都有一条多聚腺苷酸短尾的特性,设计合成了一条与其互补的多聚胸腺嘧啶苷酸,将其作为肠道病毒的通用(共用)引物,又在3种肠道病毒3′端设计合成针对某种肠道病毒的特异性引物,最后进行多重PCR。这几种引物间没有明显的交叉反应,特异性较强,可以同时检测Polio病毒、CoxB组病毒及HAV。检测细胞培养液中的病毒,其敏感性与常规PCR一致或稍低,对水中回收的病毒得到了基本相同的检测结果
Using the characteristic of a polyA tail at the 3 ’end of enterovirus (including Hepatitis A virus), a complementary polythymidine nucleotide was designed and synthesized as a universal ) Primers, and in 3 kinds of enterovirus 3 ’end designed to synthesize specific primers for a certain enterovirus, and finally multiplex PCR. There is no obvious cross-reaction between these kinds of primers, and the specificity is high. Polio virus, CoxB virus and HAV can be detected at the same time. The detection of viruses in cell culture media is consistent or slightly less sensitive than conventional PCR and yields essentially the same results for viruses recovered in water
