目的研究白藜芦醇对早期心衰模型大鼠的作用。方法 SD大鼠40只分为假手术组、模型组和白藜芦醇(10、20和40 mg·kg-1)组(均n=8),除假手术组外,其他组大鼠结扎左冠状动脉前降支制心肌梗死致早期心衰模型,制模期间灌胃给予相应剂量的溶剂或药物,共6周。采用血流动力学和心脏称重检测心功能,测定血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、一氧化氮(NO)和一氧化氮合酶(NOS)含量。结果制模6周后,模型组大鼠左心室收缩末压(LVSP)和左心室等容收缩期室内压最大上升速率(+dp/dtmax)显著下降,左心室舒张末压(LVEDP)和左心室等容舒张期室内压最大下降速率(-dp/dtmax)也显著降低,左心重/全心重均显著下降,SOD显著减少,MDA、NO和NOS显著增加,与假手术组相比有显著差异(均P<0.05);白藜芦醇20、40 mg·kg-1组大鼠LVSP和+dp/dtmax显著上升,白藜芦醇40 mg·kg-1组LVDP和-dp/dtmax显著高于模型组(P<0.05)。白藜芦醇20、40 mg·kg-1组SOD增加,MDA、NO和NOS显著降低,左心重/全心重均高于模型组(P<0.05)。讨论白藜芦醇可改善早期心衰大鼠模型的心功能,抗氧化能力增强。
Objective To study the effect of resveratrol on early heart failure model rats. Methods Forty SD rats were randomly divided into sham operation group, model group and resveratrol (10, 20 and 40 mg · kg -1) groups (all n = 8) Left anterior descending coronary artery infarction caused by early heart failure model, the model during the gavage given the appropriate dose of solvent or drug, a total of 6 weeks. Cardiac function was measured by hemodynamics and heart weight. Serum superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, nitric oxide (NO) and nitric oxide synthase (NOS) contents were measured. Results After 6 weeks of modeling, the left ventricular end-systolic pressure (LVSP) and the maximal rate of increase of ventricular pressure (+ dp / dtmax) in left ventricular systolic pressure in model group decreased significantly. Left ventricular end diastolic pressure (LVEDP) (-dp / dtmax) in ventricular diastolic phase also decreased significantly, left heart weight / total heart weight decreased significantly, SOD decreased significantly, MDA, NO and NOS increased significantly compared with sham-operated group (P <0.05). The levels of LVSP and + dp / dtmax in resveratrol 20 and 40 mg · kg-1 groups were significantly increased, and LVDP and -dp / dtmax in resveratrol 40 mg · kg-1 group were significantly increased Higher than model group (P <0.05). Resveratrol 20,40 mg · kg-1 group increased SOD, MDA, NO and NOS decreased significantly, left heart weight / total heart weight were higher than the model group (P <0.05). Discussion Resveratrol can improve the heart function of early heart failure rat model, increased antioxidant capacity.
