目的:观察栀子浸膏对Hulth法复制兔膝骨关节炎模型中软骨基质的改变及软骨与滑膜中IL-1的影响。方法:36只大耳白兔随机分为给药组、模型组和对照组,每组12只。给药组、模型组行Hulth术法复制骨关节炎模型,术后第5周,给药组以栀子浸膏600mg/kg灌胃,模型组与对照组给予等量生理盐水灌胃,给药3周后解剖显微镜下行软骨PelletierJP评分,取股骨内侧髁关节软骨行Masson及甲苯胺蓝染色观察软骨中胶原纤维、蛋白多糖含量的变化,免疫组化检测软骨与滑膜中IL-1的表达情况。结果:栀子浸膏能延缓骨关节炎关节软骨基质的丢失,给药组软骨与滑膜中IL-1的表达明显低于模型组,P<0.05。结论:栀子浸膏可减少骨关节炎模型软骨与滑膜中IL-1的表达和保护关节软骨基质,从而延缓病程的进展。
OBJECTIVE: To observe the changes of cartilage matrix and the effect of Zhizi Extract on IL-1 in cartilage and synovium with Hulth method in rabbits knee osteoarthritis model. Methods: Thirty - six large white rabbits were randomly divided into treatment group, model group and control group, with 12 rats in each group. At the fifth week after operation, the rats in the administration group were treated with Fructus Gardeniae 600mg / kg, while the rats in the model group and the control group were given the same amount of normal saline for gavage The cartilage PelletierJP score of the cartilage was measured by dissecting microscope after 3 weeks of treatment. Masson and toluidine blue staining of cartilage in medial femoral condyle were used to observe the change of collagen fibers and proteoglycan in cartilage. The expression of IL-1 in cartilage and synovium was detected by immunohistochemistry Happening. Results: Gardenia extract can delay the loss of articular cartilage in osteoarthritis. The expression of IL-1 in cartilage and synovial membrane of the treated group was significantly lower than that of the model group (P <0.05). Conclusion: Gardenia extract can reduce the expression of IL-1 in the cartilage and synovium of osteoarthritis model and protect the articular cartilage matrix, thus slowing the progression of the disease.
