目的:通过宫腔内脂质体转染的方法调控小鼠子宫内Meis1基因的表达水平,研究其对子宫内膜容受性的影响。方法:通过脂质体转染的方法在小鼠妊娠第2天向宫腔内注入Meis1相关质粒,在妊娠第4.5天观察各组小鼠子宫内膜胞饮突的表达变化。在妊娠第9天观察各组小鼠妊娠率和胚胎着床数的差异。结果:Meis1基因下调组胚胎着床数明显低于对照组;Meis1基因上调组胞饮突的发育程度高于对照组,Meis1基因下调组胞饮突的发育延后于对照组。结论:Meis1基因可能作为一种子宫内膜容受分子,在胚胎着床中发挥重要作用。
OBJECTIVE: To investigate the effect of intrauterine liposome transfection on the expression of Meis1 gene in mouse uterus and its effect on endometrial receptivity. METHODS: Meis1-related plasmids were injected into uterine cavity on day 2 of gestation in mice by liposome transfection method. The expression of Meis1 in each group was observed on day 4.5 of gestation. On the 9th day of gestation, the differences of pregnancy rate and embryo implantation number of mice in each group were observed. Results: Meis1 gene downregulation group embryo implantation number was significantly lower than the control group; Meis1 gene upregulation of the group of the development of diplopodia than the control group, Meis1 gene downregulation of the development of diplopodia in the control group. Conclusion: Meis1 gene may play an important role in embryo implantation as an endometrial receptive molecule.
