目的了解Th17细胞相关因子在人脑胶质瘤组织中的表达,确定Th17的存在及浸润程度。方法应用免疫组化二步法检测Th17细胞相关因子CD4+和IL-17在34例脑胶质瘤和10例正常脑组织病理切片中的表达,采用实时荧光定量逆转录聚合酶链反应(QRT-PCR)技术,检测IL-17mRNA、RORγtmRNA在人脑胶质瘤组织中的表达量。结果 IL-17、CD4+在正常脑组织表达不明显,在低级别脑胶质瘤中表达较少,CD4+和IL-17在高级别脑胶质瘤中表达明显,组间比较差异有统计学意义(P<0.05,P<0.05,P<0.01)。IL-17mRNA和RORγt mRNA在正常脑组织、低级别、高级别脑胶质瘤中的表达量随恶性程度升高而增加,组间比较差异有统计学意义(P<0.05,P<0.01,P<0.01)。结论脑胶质瘤中存在Th17的浸润,其表达量随脑胶质瘤恶性程度的升高而增加。
Objective To investigate the expression of Th17 cell-related factors in human glioma and to determine the existence and infiltration of Th17. Methods The expression of Th17 cell related factors CD4 + and IL-17 in 34 gliomas and 10 normal brain tissue sections were detected by immunohistochemical two-step method. Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (QRT- PCR) was used to detect the expression of IL-17 mRNA and RORγt mRNA in human glioma tissue. Results The expression of IL-17 and CD4 + in normal brain tissue was not obvious, but in low-grade gliomas, the expression of IL-17 and CD4 was significantly higher than that in high-grade glioma. The difference between the two groups was statistically significant (P <0.05, P <0.05, P <0.01). The expression of IL-17mRNA and RORγt mRNA in normal brain tissue, low-grade, high-grade glioma increased with the increase of the malignant degree, the difference between the two groups was statistically significant (P <0.05, P <0.01, P <0.01). Conclusion There is Th17 infiltration in glioma, which is increased with the malignant degree of glioma.
