目的:观察血性脑脊液对内皮细胞分泌内皮素(Endothelin,ET)和一氧化氮(Nitric Oxyde,NO)的影响。方法:建立脐静脉内皮细胞体外培养的方法,用放射免疫方法测定ET含量,用硝酸还原酶法测定NO的代谢产物NO_2~-/NO_3~-(NO_x~-)含量,并观察血性脑脊液的作用。结果:不同时间血性脑脊液均使内皮细胞ET分泌增加,NO分泌减少。结论:血性脑脊液可使体外培养的内皮细胞ET分泌增加,NO分泌减少。
Objective: To observe the effect of bloody cerebrospinal fluid (CSF) on the secretion of endothelin (ET) and nitric oxide (NO) in endothelial cells. Methods: The culture method of umbilical vein endothelial cells was established in vitro. The contents of ET were measured by radioimmunoassay. The content of NO 2 - / NO 3 - - (NO x -) was determined by nitrate reductase method and the effect of bloody cerebrospinal fluid . Results: Cerebrospinal fluid (CSF) at different times increased ET secretion and decreased NO secretion. Conclusion: Cerebrospinal fluid can increase the secretion of endotheliocyte ET in vitro and reduce the secretion of NO.
