在生物教学中,显微照片的应用极大的方便了理论教学和实验演示。常用的普通型生物显微镜配上显微摄影装置后,可以进行显微摄影。但在实际使用中却会遇到许多必须要考虑的实际问题。 1.照明问题 拍摄时可采用自然光或灯光,一般都用固定的电光源,这便于控制曝光。但在彩色摄影过程中应考虑偏色问题的出现;在用低倍物镜拍摄照片时会出现照明中像面亮度不均匀的缺点。解决办法是采用柯拉照明(即在其光源与聚光镜之间加一块毛玻璃),使得像面亮度均匀。而使用高倍物镜对此却要求不严格,因为高倍物镜的线视场小,用临界照明问题不大,稍为调整一下集光器即可使视场亮度均匀。
