[目的]观察桃红四物汤大鼠含药血清对光老化人皮肤模型成纤维细胞增殖影响与维甲酸等效性。[方法]使用随机平行对照方法,将健康雄性清洁级SD大鼠20只(体重200~250g),鼠龄4~5周使用随机数字表分为两组,10只/组;空白血清组、桃红四物汤组。使用长波紫外线照射复制光老化HSF模型。实验细胞分组:空白对照组,正常培养HSF细胞10瓶;长波紫外线照射40瓶随机分为4组,UV模型组、空白血清组、桃红四物汤血清组、维甲酸组,10瓶/组。干预培养,各组均37℃,CO2培养箱培养12h,模型组:新鲜细胞培养液5m L;桃红四物汤组:10%含药血清培养液5m L;维甲酸组:维甲酸乙酯溶液5m L;空白血清组:10%空白大鼠血清培养液5m L。连续给药干预培养12h,观测成纤维细胞增殖活性(MTT法)。[结果]成纤维细胞增殖活性各组均明显低于空白对照组(P<0.01);桃红四物汤组、维甲酸组均显著高于模型组(P<0.05),空白血清组与模型组无显著性差异(P>0.05),桃红四物汤组与维甲酸组无显著性差异(P>0.05),桃红四物汤组、维甲酸组均高于空白血清组(P<0.05)。[结论]桃红四物汤、维甲酸均可提升UVA辐射体外建立光老化人皮肤模型成纤维细胞(P<0.01),桃红四物汤与维甲酸具有等效性(P>0.05)。
[Objective] To observe the effect of Taohongsiwu Decoction-containing rat serum on the proliferation of photoreactive human skin fibroblast and the retinoic acid equivalence. [Methods] Twenty healthy male SD rats (weighing 200 ~ 250g) were randomly divided into two groups (n = 10) Taohong Siwu Tang group. Photo-Aging HSF Model Using Long-Wave Ultraviolet Irradiation. The experimental cells were divided into blank control group and normal cultured HSF cells. 40 bottles were randomly divided into 4 groups: UV model group, blank serum group, Taohongsiwutang serum group, retinoic acid group and 10 bottles / group. Intervention culture, each group were 37 ℃, CO2 incubator cultured for 12h, the model group: fresh cell culture medium 5mL; Taohongsiwutang group: 10% drug serum medium 5ml; retinoic acid group: ethyl retinoic acid solution 5m L; blank serum group: 10% blank rat serum culture solution 5m L. Continuous administration intervention for 12h, observed fibroblast proliferation activity (MTT method). [Results] The proliferation activity of fibroblasts in each group was significantly lower than that in the blank control group (P <0.01), while that in the Taohongsiwutang group and the retinoic acid group was significantly higher than that in the model group (P <0.05) There was no significant difference between the two groups (P> 0.05). There was no significant difference between Taohong Siwu Decoction group and retinoic acid group (P> 0.05). [Conclusion] Both Taohong Siwu decoction and retinoic acid can enhance UVA radiation to establish photoaging human skin fibroblasts in vitro (P <0.01), and Taohong Siwu decoction has the same effect with retinoic acid (P> 0.05).
