筛选和鉴定肝癌相关的肿瘤抗原是开展肝癌特异性免疫治疗的前提和关键.采用SEREX方法,用含有抗体的肝癌患者血清筛选肝癌cDNA表达文库,对所获得的阳性克隆进行序列测定和生物信息学分析.获得31个肝癌相关的肿瘤抗原编码基因,其中1个为未知基因.30个已知基因所编码的蛋白按功能可归为8类:肝细胞固有分子,RNA转录、剪切相关分子,蛋白质代谢相关分子,能量合成相关分子,信号转导分子,细胞黏附相关分子,免疫抑制相关分子和未知功能蛋白分子,其中包括1个CT抗原CAGE.研究表明,肝癌相关的肿瘤抗原编码基因的筛选和鉴定,为肝癌的特异性免疫治疗提供了可能靶位,并有助于肝癌的临床诊断及肝癌发生机制的理解.
Screening and identification of hepatocellular carcinoma-associated tumor antigens are the prerequisite and key to carry out hepatocellular carcinoma-specific immunotherapy.Serum cDNA of hepatocellular carcinoma was screened by serum-containing sera using SEREX method, and the positive clones were sequenced and bioinformatics 31 tumor-associated genes coding for hepatocellular carcinoma were obtained, of which 1 was unknown gene.The proteins encoded by 30 known genes could be classified into 8 types by function: hepatocyte intrinsic molecule, RNA transcription, shear related molecule, Protein metabolism related molecules, energy synthesis related molecules, signal transduction molecules, cell adhesion related molecules, immune suppression related molecules and unknown functional protein molecules, including a CT antigen CAGE .Studies show that the liver cancer-associated tumor antigen encoding gene screening And identification, provide a possible target for the specific immunotherapy of liver cancer, and contribute to the clinical diagnosis of liver cancer and understanding of the mechanism of liver cancer.
