目的 :了解化疗联合重组Endostatin对小鼠种植瘤的治疗作用 ,为肿瘤的免疫治疗提供条件。方法 :构建小鼠En dostatin原核表达质粒 ,在大肠杆菌表达小鼠Endostatin融合蛋白。分离纯化后 ,与丝裂霉素C联合应用 ,治疗种植在小鼠腿部肌肉内的小鼠肝癌H2 2肿瘤。观察瘤重 ,免疫组化检测肿瘤血管密度和肿瘤坏死情况。结论 :单纯应用重组Endostatin治疗种植瘤即产生明显的抑制作用 ,和化疗合用后 ,抑制作用更强 ,肿瘤血管密度减少 ,肿瘤细胞大量坏死。
Objective: To understand the therapeutic effect of chemotherapy combined with recombinant Endostatin on implanted tumors in mice and to provide conditions for the immunotherapy of tumors. Methods: The prokaryotic expression plasmid of mouse En dostatin was constructed and the mouse Endostatin fusion protein was expressed in E. coli. After isolation and purification, in combination with mitomycin C treatment of mouse liver cancer grown in mouse leg muscle H2 2 tumors. Observed the tumor weight, immunohistochemical detection of tumor blood vessel density and tumor necrosis. Conclusion: The treatment of implanted tumor with recombinant Endostatin alone has a significant inhibitory effect. After combined with chemotherapy, the inhibition effect is stronger, the tumor blood vessel density is reduced and a large number of tumor cells are necrotic.
