目的探讨外源性骨髓间充质干细胞(MSCs)移植对缺血/再灌注损伤(I/R)后肾小管上皮细胞凋亡以及Bcl-2、Fas和FasL表达的影响。方法将雄性SD大鼠MSCs用DAPI标记后注入受体雌性SD大鼠体内。30只受体大鼠随机分为三组(n=10):假手术对照组(C组)、MSCs+I/R组(M组)、DMEM-F12+I/R组(D组)。7d后观察肾功能、肾脏病理改变,采用原位末端标记法检测细胞凋亡指数,免疫组化法检测Bcl-2、Fas和FasL的表达,并观察DAPI标记的MSCs在受体大鼠肾脏的分布情况。结果M组在肾功能、肾脏病理改变上,均明显好于D组;M组Bcl-2蛋白表达均高于D组,而Fas、FasL蛋白表达和凋亡指数均低于D组。I/R后7d内未发现MSCs定位于肾组织中。结论外源性MSCs可以减少I/R损伤后肾小管上皮细胞凋亡,促进Bcl-2和抑制Fas、FasL表达,从而有利于肾小管损伤的早期恢复。
Objective To investigate the effect of exogenous bone marrow mesenchymal stem cells (MSCs) transplantation on the apoptosis of renal tubular epithelial cells and the expression of Bcl-2, Fas and FasL after ischemia / reperfusion injury (I / R). Methods Male SD rats were labeled with DAPI and injected into female SD rats. Thirty rats were randomly divided into three groups (n = 10): sham operation control group (C group), MSCs + I / R group (M group) and DMEM-F12 + I / R group (D group). After 7 days, the renal function and renal pathological changes were observed. The apoptotic index was detected by in situ end labeling. The expressions of Bcl-2, Fas and FasL were detected by immunohistochemistry. The expression of DAPI-labeled MSCs in the kidney of recipient rats Distribution. Results The expression of Bcl-2 protein in M group was significantly higher than that in D group, while the expression of Fas and FasL protein and apoptosis index in M group were lower than those in D group. MSCs were not found localized in the kidney within 7 days after I / R. Conclusion Exogenous MSCs can reduce the apoptosis of renal tubular epithelial cells after I / R injury, promote the expression of Bcl-2 and inhibit the expression of Fas and FasL, which is beneficial to the early recovery of renal tubular injury.
