以丰香草莓叶片为材料,MS 附加1.5 mg·L-1TDZ 和0.4 mg·L-1IBA 为基本培养基,研究了不同有色膜对草莓再生的影响,并对其机理进行了探讨。结果表明,绿膜和红膜对不定芽再生有明显的促进作用,其再生率达95%以上,平均每个外植体再生芽数25 个以上;而蓝膜和黄膜则不利于芽的分化。分析发现,不同有色膜光波差异主要集中在300~700 nm,红、绿膜在该波段光强较弱,而黄、蓝膜和荧光下较强。红膜和绿膜有较高的叶绿素含量,较低的Chl a/b 比值。抗氧化酶的活性以红膜最高,绿膜其次,黄膜最低;而丙二醛(MDA)的含量则相反。内源激素与草莓离体器官的发生之间关系密切,培养15 d 后,红、绿膜和荧光下赤霉素(GA3)的含量较高;45 d后,GA3和玉米素(ZT)的含量以红、绿膜处理最高,而吲哚乙酸(IAA)的含量以黄膜和蓝膜下的较高。
Taking Fengxiang strawberry leaves as material, MS supplemented with 1.5 mg · L-1TDZ and 0.4 mg · L-1 IBA as basic medium, the effects of different colored films on the regeneration of strawberry were studied, and the mechanism was discussed. The results showed that the green film and the red film promoted the adventitious bud regeneration obviously. After 45 days, the contents of GA3 and zeatin (ZT) The contents of red and green films were the highest, while the contents of indole acetic acid (IAA) were higher under yellow and blue films.
