目的利用超高效液相色谱配置二极管阵列检测器,旨在建立同时测定黑果腺肋花楸浆果中原花青素B1、原花青素B2、原花青素B4、芦丁和槲皮素的方法。方法色谱柱为ACQUITY UPLC?HSS T3 C18色谱柱(100 mm×2.1 mm,1.8μm);甲醇-0.03%磷酸水溶液为流动相,梯度洗脱;柱温35℃;体积流量为0.2 m L/min;检测波长:原花青素B1、原花青素B2和原花青素B4为280 nm,芦丁和槲皮素为360 nm。结果原花青素B1、原花青素B2、原花青素B4、芦丁和槲皮素分别在2.2~44.0μg/m L(r=0.999 2)、50~1 000μg/m L(r=0.999 5)、3.6~54.0μg/m L(r=0.999 2)、7.4~148.0μg/m L(r=0.999 4)和2.1~42.0μg/m L(r=0.999 7)呈良好的线性关系,平均回收率分别为101.23%、99.64%、102.31%、98.74%和103.53%,RSD分别为2.13%、1.23%、2.89%、2.96%、2.64%。黑果腺肋花楸浆果中原花青素B1、原花青素B2、原花青素B4、芦丁和槲皮素的平均质量分数分别为129.78、3 834.99、196.02、134.40、79.10μg/g。结论该方法操作简单、结果准确,具有较好的重复性和稳定性,可用于同时测定黑果腺肋花楸浆果中5种主要有效成分,将为黑果腺肋花楸质量控制提供科学依据。
Objective To establish a method for the simultaneous determination of procyanidin B1, proanthocyanidin B2, proanthocyanidin B4, rutin and quercetin in the berries of Prunus nigra L. by using ultra-performance liquid chromatography with diode array detector. METHODS The column was ACQUITY UPLC® HSS T3 C18 column (100 mm × 2.1 mm, 1.8 μm). The mobile phase consisted of methanol-0.03% phosphoric acid solution with gradient elution. The column temperature was 35 ℃ and the volume flow rate was 0.2 mL / min ; Detection wavelength: procyanidin B1, proanthocyanidin B2 and proanthocyanidin B4 280 nm, rutin and quercetin 360 nm. RESULTS: Proanthocyanidins B1, proanthocyanidins B2, proanthocyanidins B4, rutin and quercetin were in the range of 2.2 ~ 44.0 μg / mL (r = 0.999 2), 50-1 000 μg / mL (r = 0.999 4) and 2.1 ~ 42.0 μg / m L (r = 0.999 7). The average recoveries were 101.23% , 99.64%, 102.31%, 98.74% and 103.53% with RSD of 2.13%, 1.23%, 2.89%, 2.96% and 2.64%, respectively. The average contents of procyanidin B1, procyanidin B2, proanthocyanidin B4, rutin and quercetin in the berries of P. armeniaca were 129.78,3834.99,196.02,134.40 and 79.10μg / g, respectively. Conclusion The method is simple, accurate and reproducible. It can be used for the simultaneous determination of five major active components in the berry of Aronia grifola frondosa. It will provide a scientific basis for the quality control of Aronia glabra .
