【目的】利用SSR分子标记对北疆早熟陆地棉部分种质资源材料进行基因组分子检测并进行遗传多样性研究,从分子水平上揭示遗传性关系。【方法】从39个SSR核心引物中筛选多态性好且稳定的24对引物在43份北疆早熟陆地棉种质材料进行分子检测,利用NTSYS-pc 2.11软件进行遗传相似性及聚类分析。【结果】共扩增出71个多态性条带,平均每个引物为2.84个条带,扩增98个等位基因。变异的多态信息含量(PIC)在0.12~0.80。43份资源材料成对遗传相似系数变化范围在0.458~0.944,其中63.75%的材料遗传相似系数在0.500~0.696,35.71%的材料遗传相似系数在0.700~0.944。根据UPGMA聚类分析,遗传相似系数在0.58时,可将43份材料聚类为2类群。【结论】43份资源材料中大部分材料成对间相似度较高,遗传性差异较小,遗传基础较狭窄。
【Objective】 Genomic molecular detection of some germplasm resources of early-maturing upland cotton in northern Xinjiang was carried out using SSR markers and the genetic diversity was studied. The genetic relationship was revealed at the molecular level. 【Method】 Thirty-four SSR core primers were screened for 24 pairs of primers with good polymorphism and stability. The molecular markers were used to detect the germplasm resources of 43 germplasm resources of upland Upland cotton cultivars in northern Xinjiang. The genetic similarity and cluster analysis were performed using NTSYS-pc 2.11 software . 【Result】 A total of 71 polymorphic bands were amplified, with an average of 2.84 bands per primer. 98 alleles were amplified. The genetic similarity coefficient (PIC) of the polymorphic information content (PIC) between 0.12 and 0.80.43 was between 0.458 and 0.944, of which 63.75% had the genetic similarity coefficient of 0.500 ~ 0.696 and 35.71% At 0.700 ~ 0.944. According to UPGMA cluster analysis, genetic similarity coefficient of 0.58, 43 materials can be clustered into two groups. 【Conclusion】 Most of the 43 resource materials have higher similarity between pairs, smaller genetic differences and narrower genetic basis.
