目的 :研究气味沙雷氏菌粗核糖体制剂 (RP)中的抗肿瘤活性成分.方法 :以RP的多克隆抗血清做Western blot ,寻找制剂中特异性的免疫原 ;以DEAE 32离子交换层析法结合SephadexG 2 0 0层析法纯化之 ,体外细胞毒试验快速鉴定其抑瘤活性.结果 :RP及盐洗核糖体制剂 (SR)有两种特异性的免疫原 ,分子量分别为 16 .5kDa和 12kDa ,而层析核糖体制剂 (CR)中却没有 ;纯化的 16 .5kDa蛋白质印渍反应与RP中 16 .5kDa处的印渍反应一致 ;体外细胞毒性试验发现它对P388细胞的生长有抑制作用 ,IC50 可达 0 .0 0 1g/L ,但它不影响WISH细胞的生长.结论 :分子量为 16 .5kDa蛋白质是气味沙雷氏菌核糖体制剂体外抑瘤作用的有效成分.“,”Objective: To explore the antitumor components of S. odorifera.Methods: Specific immunogen of S. odorifera were identified with multicloned antiserum of RP by Western blot, and were seperated and purified by DEAE ion changed chromatograph combined with Sephadex G 200 chromatograph. Results: Experiments proved that a 16.5 kDa protein and a 12 kDa protein are the specific immunogen of S. odorifera. The 16.5 kDa protein was seperated and purified, and this protein is cytotoxic to P 388 cell lines. Conclusion: The 16.5 kDa protein is an effective component of S. odorifera with antitumor activity in vitro.
