目的:探讨脆性组氨酸三联体(fragile histidine trail,FHIT) 蛋白在45例结直肠癌、36例结直肠腺瘤和23例正常对照组中的表达睛况. 方法:标本经40 g/L甲醛固定,常规石蜡包埋,采用免疫组化SP法检测FHIT在结直肠癌、结直肠腺瘤和正常对照组中的表达. 结果:在45例结直肠癌中28例FHIT蛋白表达显著减少或缺失,5例表达减弱,12例表达阳性;其中结直肠癌Dukes A,B和C期FHIT蛋白表达阴性比例分别为6/11,5/12 和17/22.在36例结直肠腺瘤中7例FHIT蛋白表达显著减少或缺失,4例表达减弱,25例表达阳性;其中结直肠腺瘤伴轻、中和重度不典型增生FHIT蛋白表达阴性比例分别为2/16,1/7和4/13.FHIT蛋白阴性表达在结直肠癌和正常对照组(62.2% vs 4.4%,X2=25.33,P<0.01)、结直肠癌和结直肠腺瘤(62.2% vs 19.4%,X2=14.19,P<0.01)、结直肠癌未伴淋巴转移的Dukes A+B期和已伴淋巴转移的C 期(47.8% vs 77.3%,X2=4.28,P<0.05)间均有显著性差异,而在结直肠腺瘤伴轻度不典型增生和中、重度不典型增生(12.5% vs 25.0%,X2=0.84,P>0.25),结直肠腺瘤和正常对照组(19.4% vs 4.4%,X2=3.51,P>0.05)中无显著性差异. 结论:FHIT蛋白在结直肠癌中呈低表达或缺失,且FHIT低表达与转移状况有相关关系,提示该基因可能与结直肠癌的发生发展有关.
Objective: To investigate the expression of fragile histidine trail (FHIT) protein in 45 cases of colorectal cancer, 36 cases of colorectal adenoma and 23 cases of normal control.Methods: Formaldehyde fixation and routine paraffin embedding were used to detect the expression of FHIT in colorectal cancer, colorectal adenoma and normal control group by immunohistochemical SP method.Results: The expression of FHIT protein in 28 cases of 45 cases of colorectal cancer was significantly decreased or 5 cases were weakened and 12 cases were positive. The negative expression rates of FHIT protein in colorectal cancer Dukes A, B and C were 6/11, 5/12 and 17/22, respectively. In 36 colorectal adenomas The expression of FHIT in 7 cases was significantly reduced or absent, and the expression of FHIT in 4 cases was weakened and 25 cases were positive. The negative expression rate of FHIT protein in colorectal adenoma with 2 cases, 2 cases, 7 cases and 4 cases /13.FHIT protein negative expression in colorectal cancer and normal control group (62.2% vs 4.4%, X2 = 25.33, P <0.01), colorectal cancer and colorectal adenoma (62.2% vs 19.4%, X2 = 14.19, P <0.01). There was significant difference between Dukes A + B and non-lymph node metastasis C (47.8% vs 77.3%, X2 = 4.28, P <0.05) (12.5% vs 25.0%, X2 = 0.84, P> 0.25), colorectal adenoma and normal control group (19.4% vs 4.4 %, X2 = 3.51, P> 0.05) .Conclusion: FHIT protein is low or absent in colorectal cancer, and low correlation between FHIT expression and metastasis suggests that this gene may be associated with colorectal cancer Related to the occurrence and development.
