目的 系统研究正常SD大鼠各组织内钠泵α亚单位三种异构体的分布情况。方法 采用分子生物学RT PCR及免疫组织化学技术 ,检测正常大鼠多种组织内钠泵α1、α2及α3亚单位mRNA及蛋白水平的表达。结果 左室心肌主要表达α1与α2亚单位 ,α3亚单位表达较弱 ;肝脏中在mRNA水平α1亚单位表达较强 ,α2与α3亚单位较弱 ,而在蛋白水平三者表达均较弱 ;肾脏中主要表达α1亚单位 ,α2与α3亚单位较弱 ;肾上腺中α1亚单位的表达略高于α2与α3亚单位 ;动脉平滑肌中α2亚单位的表达略高于α1与α3亚单位 ;垂体中三种亚单位的表达均较强 ;下丘脑中α2与α3亚单位表达较强 ,α1亚单位表达较弱。结论 本研究系统揭示了正常大鼠多种组织内钠泵α亚单位三种异构体在mRNA及蛋白水平的表达情况 ,为钠泵生理及病理作用的进一步研究提供了理论及实验依据
Objective To study the distribution of three isoforms of sodium pump α subunit in normal SD rats. Methods The mRNA and protein expressions of α1, α2 and α3 subunits of sodium pump in various tissues of normal rats were detected by molecular biological RT-PCR and immunohistochemistry. Results The expression of α1 and α2 subunits and the expression of α3 subunit in left ventricular myocardium were weak. The expression of α1 subunit mRNA, α2 and α3 subunits were weak in liver and weak in the protein level. Α1 and α3 subunits were weakly expressed in the kidneys. The expressions of α1 subunits in the adrenal glands were slightly higher than those in α2 and α3 subunits. The expression of α2 subunits in arterial smooth muscle was slightly higher than that of α1 and α3 subunits. Conclusions The present study revealed the expression of three isoforms of sodium pump α subunit in various tissues of normal rats at mRNA and protein levels, providing theoretical and experimental evidence for the further study of the physiological and pathological effects of sodium pump
