目的:观察通心络(Tongxinluo,TXL)超细干粉水提物对异丙肾上腺素(isoproterenol,Iso)诱导的H9c2心肌细胞肥大的作用,并探讨其对缝隙连接蛋白43(connexin 43,Cx43)mRNA表达的影响。方法:将培养的H9c2心肌细胞随机分为对照组、TXL组、Iso组和Iso+TXL组,通心络以通心络超细干粉水提物的形式加入,使用异丙肾上腺素诱导72 h后,采用相差显微镜测量细胞大小,BCA法测定蛋白浓度,运用逆转录聚合酶链式反应(RT-PCR)测定Cx43mRNA的表达。结果:Iso刺激H9c2心肌细胞72 h后,Iso组细胞直径增大,高于对照组(P<0.05);通心络对正常H9c2心肌细胞直径无明显影响,但能抑制Iso诱导的细胞直径增大(P<0.05)。Iso组蛋白浓度明显升高,高于对照组(P<0.05);通心络对正常H9c2心肌细胞蛋白浓度无明显影响,但能抑制Iso诱导的蛋白浓度的升高(P<0.05)。Iso组Cx43mRNA表达明显减少,低于对照组(P<0.05);通心络对正常H9c2心肌细胞Cx43mRNA表达无影响,但能够抑制Iso诱导的Cx43 mRNA表达的下调(P<0.05)。结论:通心络可以抑制Iso诱导的H9c2心肌细胞肥大及Cx43mRNA表达的下调。
OBJECTIVE: To observe the effects of Tongxinluo (TXL) ultrafine water extract on isoproterenol (Iso) -induced H9c2 cardiomyocyte hypertrophy and to explore its effect on the expression of connexin 43 (Cx43) Effect of mRNA expression. Methods: The cultured H9c2 cardiomyocytes were randomly divided into control group, TXL group, Iso group and Iso + TXL group. Tongxinluo was added into Tongxinluo superfine dry powder water extract, and isoproterenol was used to induce 72 h The cell size was measured by phase-contrast microscopy. The protein concentration was determined by BCA method. The expression of Cx43 mRNA was determined by reverse transcriptase-polymerase chain reaction (RT-PCR). RESULTS: After Iso stimulation of H9c2 cardiomyocytes for 72 h, the diameter of cells in Iso group increased significantly (P <0.05). Tongxinluo had no effect on the diameter of normal H9c2 cardiomyocytes but inhibited the increase of Iso-induced cell diameter Large (P <0.05). Iso group protein concentration increased significantly higher than the control group (P <0.05); Tongxinluo no effect on normal H9c2 myocardial cell protein concentration, but can inhibit Iso-induced protein concentration increased (P <0.05). Compared with control group, the expression of Cx43 mRNA in Iso group was significantly lower than that in control group (P <0.05). Tongxinluo had no effect on the expression of Cx43 mRNA in normal H9c2 cardiomyocytes but decreased the Iso-induced Cx43 mRNA expression (P <0.05). Conclusion: Tongxinluo can inhibit Iso-induced H9c2 cardiomyocyte hypertrophy and down-regulation of Cx43 mRNA expression.
