近二十年以来,大量研究工作表明,同一药物,同一剂量以及相同的给药途径,由于个体差异和一些其它因素的影响,在作用部位或受体反应部位的药物浓度可有显著差别。例如南京医学院第一附属医院对以常规剂量给药的苯妥英钠等四种药物的有效、无效、中毒百分率及相应血药浓度作如下比较。从上表看出,如单纯按照常规剂量给药,对很多患者无效,但有的患者却有中毒的可能。这种在常规用量时某些患者的疗效很小,而另一些患者则可引起严重中毒的现象,仍是长期以来临床医师和药理学家所共同关心的问题之一。
