目的观察雷公藤多苷(TWP)对糖尿病肾病(DN)大鼠核因子κB(NF-κB)、C-C型趋化因子配体5(CCL5)、肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)等炎性细胞因子表达的影响。方法链脲佐菌素(STZ)腹腔注射建立DN大鼠模型。动物随机分为正常组、模型组、1.8 g/kg/d TWP治疗组。用药8周末,计算肾脏指数(KI)、全自动生化分析仪检测大鼠血糖(BG)、糖化血红蛋白(HbA1c)、血肌酐(Scr)、血尿素氮(BUN)等生化指标。ELISA检测大鼠血清超敏C反应蛋白(hs-CRP)和肾匀浆IL-6、TNF-α和CCL5的含量;HE染色观察大鼠肾脏病理学改变;免疫组织化学染色检测大鼠肾组织NF-κB的表达。结果与模型组相比,TWP可改善DN大鼠一般状况;明显降低DN大鼠空腹BG、HbA1c、KI、BUN及Scr水平,增加DN大鼠体质量(P<0.01);与模型组比较,TWP明显降低DN大鼠血清hs-CRP水平及肾匀浆中IL-6、TNF-α和CCL5的水平,明显下调DN大鼠肾组织NF-κB的表达(P<0.01),经TWP治疗后DN大鼠肾脏病理变化减轻。结论 TWP可降低糖尿病大鼠血清及肾脏炎性细胞因子表达,减轻肾脏病变。
Objective To observe the effects of Tripterygium wilfordii glycosides (TWP) on NF-κB, CCL5, TNF-α and leukocyte in rats with diabetic nephropathy (DN) 6 (IL-6) and other inflammatory cytokines expression. Methods DN rats were established by intraperitoneal injection of streptozotocin (STZ). Animals were randomly divided into normal group, model group, 1.8 g / kg / d TWP treatment group. After 8 weeks of treatment, the kidney index (KI) and the biochemical indexes such as blood glucose (BG), HbA1c, serum creatinine (BUN) and blood urea nitrogen were detected by automatic biochemical analyzer. The contents of hs-CRP and IL-6, TNF-α and CCL5 in serum were detected by ELISA. The pathological changes of rat kidney were observed by HE staining. The expressions of hs-CRP, NF-κB expression. Results Compared with the model group, TWP could improve the general condition of DN rats, significantly lower the fasting BG, HbA1c, KI, BUN and Scr levels and increase the body weight of DN rats (P <0.01); Compared with the model group, TWP significantly reduced the level of hs-CRP in serum and the levels of IL-6, TNF-α and CCL5 in renal homogenate, and significantly decreased the expression of NF-κB in DN rats (P <0.01) DN rat kidney pathological changes reduced. Conclusion TWP can reduce the expression of inflammatory cytokines in serum and kidneys of diabetic rats and reduce the renal lesions.
