本实验主要目的是将从β地贫患者DNA中制得的大小约2.6kb含β珠蛋白基因的基因组片段,克隆到载体M13mp19上,筛选出含β基因的重组体,用于核苷酸序列分析,以发现中国人β地贫珠蛋白基因的突变位点,进一步探讨β地贫发病的分子基础。实验所用主要方法有基因的分离提取,连接转化,杂交筛选。实验结果未能得到所需的重组体。
The main purpose of this experiment was to clone the genome fragment containing the β-globin gene, about 2.6 kb in size, from the DNA of patients with β thalassemia into the vector M13mp19. The recombinant plasmid containing the β gene was screened for the nucleotide sequence Analysis, in order to find the Chinese human β-thalassemia gene mutation sites, to further explore the molecular basis of β thalassemia. The main methods used in the experiment were gene isolation and extraction, transformation and hybridization screening. Experimental results failed to obtain the desired recombinant.
