目的 :探讨重组人转化生长因子 β3 (rhTGFβ3 )在伤口愈合及瘢痕形成中的作用机制。方法 :取体外培养的人正常皮肤和增生性瘢痕成纤维细胞 (normalskinfibroblast,NSFb ;hypertrophicscarfibroblast ,HSFb) ,应用不同浓度rhTGFβ3 ,通过放射免疫和Northernbloting杂交方法观察NSFb和HSFbⅠ、Ⅲ型胶原蛋白及转录水平mRNA表达的变化。结果 :(1)人正常皮肤和瘢痕组织成纤维细胞Ⅰ、Ⅲ型前胶原有明显的不同表达 ;(2 )与对照组相比 ,实验组 (分别加rhTGFβ3 ,终浓度为 1、5、10ng·L-1)Ⅰ、Ⅲ前胶原合成明显增加 (P <0 .0 0 1) ,Ⅰ /Ⅲ前胶原的比例变小 ;(3 )实验组rhTGFβ3 对NSFb和HSFb生物学作用的影响呈明显的量效关系。结论 :rhTGFβ3 可有效提高成纤维细胞Ⅰ、Ⅲ型胶原的合成和Ⅰ、Ⅲ型构成比中Ⅲ型胶原的相对含量 ,其对加速创面愈合及减少瘢痕形成可能具有重要作用
Objective: To investigate the mechanism of recombinant human transforming growth factor β3 (rhTGFβ3) in wound healing and scar formation. Methods: Human normal skin and hypertrophic scar fibroblasts (NSFb) were cultured in vitro. The expressions of NSFb, HSFb Ⅰ, Ⅲ collagen and transcription level were detected by radioimmunoassay and Northern blotting with different concentrations of rhTGFβ3. Changes in mRNA expression. Results: (1) The expression of type Ⅰ procollagen and type Ⅲ procollagen in normal human skin and scar tissue were significantly different. (2) Compared with the control group, the experimental group (rhTGFβ3, final concentration of 1, (P <0.01), the ratio of Ⅰ / Ⅲ procollagen became smaller; (3) The effect of rhTGFβ3 in the experimental group on the biological effects of NSFb and HSFb was obvious The dose-effect relationship. CONCLUSION: rhTGFβ3 can effectively increase the synthesis of type Ⅰ and Ⅲ fibroblasts and the relative content of type Ⅲ and type Ⅲ collagen in fibroblasts, which may play an important role in accelerating wound healing and reducing scar formation
