目的:探讨食管鳞癌组织中细胞周期素B1和D1(Cyclin B1,Cyclin D1)mRNA和蛋白的表达及其与食管鳞癌床生物学指标的关系.方法:应用原位杂交法、免疫组化SP法对62例食管鳞癌组织(组织学Ⅰ级15例,Ⅱ级25例,Ⅲ级22例;有淋巴结转移20例,无淋巴结转移42例;肿瘤浸润深度:浸润至黏膜层、黏膜下层或浅肌层7例,深肌层14例,纤维膜41例)、31例癌旁不典型增生组织及62例正常食管黏膜组织进行Cyclin B1,D1 mRNA和蛋白的检测,分析其阳性表达与食管鳞癌患者临床病理因素的关系.结果:Cyclin B1,D1的mRNA和蛋白在正常食管黏膜组织中不表达或低表达,在癌旁不典型增生组织及食管鳞癌组织中高表达,三者阳性表达率比较差异有统计学意义(P<0.05);食管鳞癌患者癌组织中Cyclin B1,D1的mRNA和蛋白的表达与性别、年龄无关;与组织学分级、浸润深度及有无淋巴结转移有关(P<0.05).Cyclin B1,D1 mRNA与蛋白正常食管黏膜组织、癌旁不典型增生组织及食管鳞癌组织中的表达有高度的一致性.结论:CyclinB1,D1 mRNA和蛋白的高表达可促进食管鳞癌的发生与发展,在癌前病变期的鳞状上皮不典型增生组织中即出现高表达,是食管鳞癌发生过程中的早期事件.
Objective: To investigate the expression of Cyclin B1 and Cyclin D1 mRNA and protein in esophageal squamous cell carcinoma and its relationship with the biological indicators of esophageal squamous cell carcinoma.Methods: The in situ hybridization, immunohistochemistry SP in 62 cases of esophageal squamous cell carcinoma (histological grade Ⅰ in 15 cases, Ⅱ grade in 25 cases, Ⅲ grade in 22 cases; lymph node metastasis in 20 cases, no lymph node metastasis in 42 cases; tumor invasion depth: infiltration to the mucosa, submucosa 7 cases of superficial myometrial muscle, 14 cases of deep myometrium and 41 cases of fibrous membrane), 31 cases of atypical hyperplasia of cancerous tissues and 62 cases of normal esophageal mucosa. The expressions of Cyclin B1, D1 mRNA and protein were detected, Esophageal squamous cell carcinoma patients.Results: Cyclin B1, D1 mRNA and protein in normal esophageal mucosa tissue is not expressed or low expression in the adjacent atypical hyperplasia and esophageal squamous cell carcinoma tissues were highly expressed, the three were positive (P <0.05). The expression of Cyclin B1 and D1 mRNA and protein in esophageal squamous cell carcinoma was not related to gender and age, and was related to histological grade, depth of invasion and lymph node metastasis (P <0.05) .Cyclin B1, D1 mRNA and protein were positive Esophageal mucosa, paraneoplastic atypical hyperplasia and esophageal squamous cell carcinoma.Conclusion: High expression of CyclinB1, D1 mRNA and protein can promote the occurrence and development of esophageal squamous cell carcinoma, Of squamous cell atypical hyperplasia of tissue that is the high expression of esophageal squamous cell carcinoma is an early event in the process.
