目的:研究白杨素(chrysin,ChR)抑制肺癌A549细胞系球形成细胞自我更新作用。方法:不同浓度ChR处理体外培养A549细胞系球形成细胞。肿瘤球形成法测定自我更新。Western blot分析CK2α和Gli1蛋白表达。pcDNA3.1-CK2α转染探讨作用机制。结果:ChR(10、20和40μM)处理24小时显著降低A549细胞系肿瘤球形成率(P<0.05),并伴随着CK2α和Gli1蛋白表达下调。与pcDNA3.1-LacZ转染细胞相比,pcDNA3.1-CK2α转染A549细胞系球形成细胞CK2α和Gli1蛋白表达显著上调,并增高肿瘤球形成率和减弱ChR的作用。结论:下调CK2α和Gli1表达参与ChR抑制A549细胞系球形成细胞自我更新作用。
OBJECTIVE: To study the effect of chrysin (ChR) on the self-renewal of human lung cell line A549 cells. Methods: Splenocytes of A549 cell line were cultured in vitro with different concentrations of ChR. Tumor formation assay self-renewal. Western blot analysis CK2α and Gli1 protein expression. The mechanism of pcDNA3.1-CK2α transfection was explored. Results: ChR (10, 20 and 40μM) for 24 hours significantly reduced the rate of tumor formation in A549 cells (P <0.05), accompanied by the down-regulation of CK2α and Gli1 protein expression. Compared with pcDNA3.1-LacZ transfected cells, pcDNA3.1-CK2α transfected A549 cells significantly increased the expression of CK2α and Gli1 protein, increased the rate of tumor formation and decreased the role of ChR. CONCLUSION: Down-regulation of CK2α and Gli1 expression is involved in the effect of ChR on self-renewal of spherical cell line A549.
