目的 观察 1,8-二羟基 - 3,5 -二甲氧基酮 (XT)对反复脑缺血再灌后脑内氨基酸及乙酰胆碱酯酶活力的影响。方法 手术前给小鼠 iv XT5 0和 10 m g/ kg,对反复脑缺血再灌注后 45 min,用氨基酸自动分析仪测定小鼠脑内谷氨酸 (Glu) ,天冬氨酸 (Asp)和 γ-氨基丁酸 (GABA)的含量 ;并测定造模后 45 m in以及第 5天脑内乙酰胆碱酯酶 (Ach E)活力。结果 反复脑缺血再灌注后 45 min,Glu和 Asp均升高 ,GABA也随之升高。二个剂量均可抑制Glu、Asp的升高 ,5 0 mg/ kg XT还使升高的 GABA恢复至接近正常。造模后 45 m in以及第 5天 ,模型组小鼠 Ach E活力降低 ,XT可升高 Ach E活力。结论 XT可抑制反复脑缺血再灌后脑内 Glu,Asp和 GABA含量的升高 ,提高Ach E活力
Objective To observe the effects of 1,8-dihydroxy-3,5-dimethoxycholine (XT) on brain amino acids and acetylcholinesterase (AChE) activity after repeated cerebral ischemia and reperfusion in rats. Methods The mice were anesthetized with iv XT50 and 10 mg / kg, and the contents of glutamic acid (Glu), aspartic acid (Asp) were determined by automatic amino acid analyzer after 45 min of repeated cerebral ischemia-reperfusion Γ-aminobutyric acid (GABA) were measured. The brain acetylcholinesterase (AchE) activity was measured 45 min after modeling and on the 5th day. Results After 45 min of repeated cerebral ischemia-reperfusion, both Glu and Asp increased and GABA also increased. Both doses inhibited both Glu and Asp, and 50 mg / kg XT also restored elevated GABA to near normal. At 45 m in and 5 d after model establishment, the Ach E activity of model mice decreased, while XT increased Ach E activity. Conclusion XT can inhibit the increase of Glu, Asp and GABA contents in the brain after repeated cerebral ischemia and reperfusion, and improve the activity of Ach E
