目的:建立MCF-10AT乳腺癌癌前病变模型,探讨复方仙蓉颗粒(Compound Xian Rong Granules,CXRG)对MCF-10AT乳腺癌癌前病变模型的抑制作用。方法:采用体外接种MCF-10AT细胞于BALB/c雌性裸鼠,并肌注苯甲酸雌二醇建立乳腺癌癌前病变模型。随机分为模型组、CXRG组、三苯氧胺(Tamoxifen,TAM)组,每组20只。分接种后21、35、49、63天,分组观察裸鼠移植瘤重量及体积变化,观察移植瘤组织形态学变化。结果:接种后21天起裸鼠移植瘤组织均出现不同程度的增生。接种后35~49天,3组裸鼠移植瘤组织均出现不同程度的不典型增生,发生率为80%~100%。接种后63天,共出现3例浸润癌,其中模型组2/7,TAM组1/7;而CXRG组表现与49天时相似。结论:复方仙蓉颗粒能抑制或减缓MCF-10AT乳腺癌癌前病变的增殖,作用优于三苯氧胺。
OBJECTIVE: To establish a model of MCF-10AT breast cancer precancerous lesion, and to investigate the inhibitory effect of Compound Xian Rong Granules (CXRG) on MCF-10AT breast cancer precancerous lesion. Methods: MCF-10AT cells were inoculated into BALB / c female nude mice and the model of breast precancerous lesions was established by intramuscular injection of estradiol benzoate. Randomly divided into model group, CXRG group, Tamoxifen (TAM) group, 20 rats in each group. At 21, 35, 49, and 63 days after the inoculation, the weight and volume changes of the transplanted tumor in nude mice were observed by grouping and the morphological changes of the transplanted tumor were observed. Results: The nude mice xenografted tumor tissues appeared different degrees of proliferation 21 days after inoculation. 35 to 49 days after inoculation, three groups of nude mice transplantation showed varying degrees of atypical hyperplasia, the incidence was 80% to 100%. Sixty-three days after inoculation, there were 3 cases of invasive carcinoma, of which 2/7 in the model group and 1/7 in the TAM group. The performance of the CXRG group was similar to that of the 49-day group. CONCLUSION: Fufang Xianrong Granule can inhibit or slow the proliferation of MCF-10AT breast cancer precancerous lesion, which is better than tamoxifen.
