富含脯氨酸的酪氨酸激酶2(proline-rich tyrsosine kinase 2,Pyk2)是一种非受体型酪氨酸激酶,与黏着斑激酶(focal adhesion kinase,FAK)共同组成黏着斑激酶家族。Pyk2在多种器官及组织中表达,受一系列因子激活,参与多条信号通路的信息传递,能够调节细胞的迁移、增殖、分化及凋亡等。在许多肿瘤中均发现Pyk2的异常表达,因此Pyk2可能成为肿瘤治疗的新靶点。本文就Pyk2的结构与分布、在多种肿瘤(骨髓瘤、乳腺癌、前列腺癌、卵巢癌)中的调控作用及其抑制剂的应用方面作一综述。
Proline-rich tyrosine kinase 2 (Pyk2) is a non-receptor tyrosine kinase that forms a family of focal adhesion kinases together with focal adhesion kinase (FAK). . Pyk2 is expressed in various organs and tissues, activated by a series of factors, and involved in the information transmission of multiple signaling pathways, which can regulate cell migration, proliferation, differentiation, and apoptosis. Abnormal expression of Pyk2 is found in many tumors, so Pyk2 may become a new target for tumor treatment. This article reviews the structure and distribution of Pyk2, its regulation in various tumors (myeloma, breast cancer, prostate cancer, and ovarian cancer) and its application in the field of inhibitors.
