目的:研究肌醇六磷酸(IP6)对HT-29细胞裸鼠皮下移植瘤生长的抑制作用。方法:建立HT-29细胞裸鼠移植瘤模型,随机分为对照组(生理盐水组)、低剂量IP6组和高剂量IP6组,用药20d后处死小鼠。比较各组抑瘤作用和裸鼠一般状态的变化,透射电镜观察移植瘤细胞形体变化,TUNEL法检测移植瘤细胞凋亡情况,并运用RT-PCR、蛋白质印迹法检测移植瘤Bcl-2及Bax基因和蛋白的表达。结果:IP6组出现典型凋亡现象,PCR及蛋白检测也显示IP6可显著上调Bax蛋白的表达,下调Bcl-2蛋白的表达(P<0.05);在移植瘤质量和体积方面,用药组与对照组差异无统计意义,P>0.05,但用药组移植瘤呈现下降趋势。结论:IP6可能通过促进细胞凋亡而起到抑制和延缓肿瘤生长的作用,其机制可能与上调Bax蛋白表达和下调Bcl-2蛋白表达有关。
Objective: To study the inhibitory effect of phytate (IP6) on the growth of subcutaneously transplanted HT-29 cells in nude mice. Methods: The transplanted model of HT-29 cells in nude mice was established and randomly divided into control group (saline group), low dose IP6 group and high dose IP6 group. The mice were sacrificed 20 days later. The antitumor effects and the general state of nude mice in each group were compared. The morphological changes of the transplanted tumor cells were observed by transmission electron microscopy. The apoptosis of the transplanted tumor cells was detected by TUNEL. The expressions of Bcl-2 and Bax in the transplanted tumor were detected by RT-PCR and Western blotting Gene and protein expression. Results: Apoptosis was observed in IP6 group. PCR and protein detection also showed that IP6 significantly up-regulated the expression of Bax protein and down-regulated the expression of Bcl-2 protein (P <0.05). In terms of the quality and volume of transplanted tumor, Group differences were not statistically significant, P> 0.05, but the treatment group showed a downward trend in xenografts. Conclusion: IP6 may play a role in the inhibition and delay of tumor growth by promoting apoptosis. The mechanism may be related to up-regulation of Bax protein expression and down-regulation of Bcl-2 protein expression.
