目的探讨信号传导和转录激活因子1(STAT1)及其抑制因子PIAS1与生殖道人乳头瘤病毒(HPV)感染及宫颈鳞癌的关系。方法采取免疫组化SABC法检测湘雅三医院及湘雅二医院2003年6月至2007年10月病理科存档的正常宫颈、宫颈上皮内瘤变(CIN)、Ⅰ期及Ⅱ期宫颈鳞癌组织石蜡块中STAT1及PIAS1的表达。结果 (1)STAT1及PIAS1在不同宫颈组织中均有表达。在正常宫颈组、CIN组与宫颈鳞癌组中,STAT1表达依次减弱,而PIAS1的表达依次增强,差异均有统计学意义(P<0.01),三组间两两比较差异亦均有统计学意义(P<0.01),且两者表达呈明显负相关(P<0.01)。(2)正常宫颈组中,STAT1及PIAS1表达HPV阳性组均显著强于HPV阴性组(P<0.05)。(3)在宫颈鳞癌组内,STAT1及PIAS1表达与肿瘤的FIGO分期、淋巴结转移及病理学分级有关。结论生殖道HPV感染可能导致STAT1及PIAS1的异常表达,同时两者异常表达可能与宫颈癌前病变及宫颈鳞癌发生、发展有关。
Objective To investigate the relationship between signal transducers and activators of transcription 1 (STAT1) and its inhibitor PIAS1 and genital human papillomavirus (HPV) infection and cervical squamous cell carcinoma. Methods Immunohistochemical SABC method was used to detect the expressions of normal cervical and cervical intraepithelial neoplasia (CIN), stage Ⅰ and Ⅱ cervical squamous cell carcinoma of the Xiangya Third Hospital and Xiangya Second Hospital from June 2003 to October 2007, Tissue paraffin block expression of STAT1 and PIAS1. Results (1) STAT1 and PIAS1 were expressed in different cervical tissues. In normal cervix group, CIN group and cervical squamous cell carcinoma group, the expression of STAT1 gradually decreased, while the expression of PIAS1 increased in turn, with statistical significance (P <0.01). The difference between the two groups was also statistically significant (P <0.01), and there was a significant negative correlation between the two expressions (P <0.01). (2) In the normal cervix group, the expression of STAT1 and PIAS1 in HPV positive group were significantly stronger than that in HPV negative group (P <0.05). (3) In the cervical squamous cell carcinoma group, the expression of STAT1 and PIAS1 correlated with FIGO stage, lymph node metastasis and pathological grade. Conclusions Genital tract HPV infection may lead to the abnormal expression of STAT1 and PIAS1, and the abnormal expression of both may be related to the development of cervical precancerous lesions and cervical squamous cell carcinoma.
