目的:探讨糖尿病大鼠睾丸组织黄嘌呤氧化酶(XOD)、髓过氧化物酶(MPO)和线粒体琥珀酸脱氢酶(SDH)的变化及灵芝孢子粉对睾丸组织的保护作用。方法:将清洁级Wistar大鼠50只,随机分成3组(对照组10只、模型组20只、灵芝孢子粉组20只)。模型组及灵芝孢子粉组以25mg/kg的剂量一次性腹腔注射2%链脲佐菌素,对照组腹腔注射等量柠檬酸钠-柠檬酸缓冲液。正常饮食喂养2周后,进行糖耐量试验,模型组和灵芝孢子粉组糖耐量异常者保留,余去除,并改喂高脂高糖饮食,灵芝孢子粉组另加灵芝孢子粉[250mg/(kg.d)]持续10周,动物均单独喂养,实验结束前1d做糖耐量试验,取睾丸。检测睾丸组织XOD、MPO和线粒体SDH。结果:模型组睾丸组织的SDH较对照组和灵芝孢子粉组明显降低(P<0.05),而XOD和MPO明显高于对照组和灵芝孢子粉组(P<0.05)。模型组生精小管层次不清,精子生成量减少或消失,管腔不规则,腔周围纤维组织(间质)增生,基膜增厚,血管壁纤维组织样增生硬化。灵芝孢子粉组生精小管层次比较清析,管腔规则,有大量精子生成,间质无增生,血管基膜增生硬化不明显。结论:灵芝孢子粉能减少自由基对睾丸组织的损伤,提高SDH活性,从而对糖尿病大鼠睾丸组织起保护作用。
Objective: To investigate the changes of xanthine oxidase (XOD), myeloperoxidase (MPO) and mitochondrial succinate dehydrogenase (SDH) in the testis of diabetic rats and the protective effect of Ganoderma lucidum spores on testicular tissue. Methods: Fifty Wistar rats of clean grade were randomly divided into three groups (10 in the control group, 20 in the model group, and 20 in the Ganoderma lucidum spore powder group). The model group and the Ganoderma lucidum spore powder group were intraperitoneally injected with 2% streptozotocin at a dose of 25 mg/kg, and the control group was intraperitoneally injected with the same amount of sodium citrate-citrate buffer. After 2 weeks of normal diet, glucose tolerance test was performed. The model group and the ganoderma lucidum spore powder group had abnormal glucose tolerance and were removed, and were fed a high fat and high sugar diet. Ganoderma lucidum spore powder group was added with Ganoderma lucidum spore powder [250mg/( Kg.d)] For 10 weeks, the animals were fed separately. The glucose tolerance test was performed 1 d before the end of the experiment and the testes were taken. Testicular tissue XOD, MPO and mitochondrial SDH were examined. Results: Compared with the control group and the Ganoderma lucidum spore powder group, the SDH of the testis tissue in the model group was significantly lower (P<0.05), while the XOD and MPO were significantly higher than the control group and the Ganoderma lucidum spore powder group (P<0.05). In the model group, the level of seminiferous tubules was unclear, sperm production decreased or disappeared, lumens were irregular, fibrosis (stroma) around the lumen proliferated, basement membrane thickened, and fibrous tissue-like proliferation and sclerosis of the vascular wall. In the Ganoderma lucidum spore powder group, the level of seminiferous tubules was relatively clear, the lumen was regular, there was a large amount of spermatozoa, and there was no hyperplasia in the stroma, and the proliferation and hardening of the vascular basement membrane was not obvious. Conclusion: Ganoderma lucidum spore powder can reduce the free radical damage to testicular tissue, increase the activity of SDH, and thus protect the testis of diabetic rats.
