为建立鲉形目内各物种的快速鉴别方法,本研究扩增了我国47个鲉形目物种并下载了Gen Bank上收录的鲉形目共计23科85属233种873条细胞色素C氧化酶I基因(COI)序列,分析该基因结构特性。结果表明,鲉形目DNA条形码基因的碱基变异中,不变位点508个,占总位点数的82.1%;转换位点70个,颠换位点41个,分别占总位点数的11.3%和6.6%;种内遗传距离为0~0.019,平均遗传距离为0.003?0.0034;属内种间遗传距离为0.003~0.258,平均值为0.086?0.038;基于233个物种的COI序列构建的系统进化树显示,227个物种(97.4%)能聚为独立分支,且具有较高支持度。在此基础上以鲉科11属39种鱼类的DNA条形码为例,筛选出25个种的37个特异性探针,以此探针对鲉科鱼类进行物种快速鉴定成功率为64.1%,研究结果表明DNA条形码芯片用于鲉科的鉴定具有一定的可行性。
In order to establish a rapid identification method for each species in Castanoplastrum, 47 species of Castanopsis species in China were amplified and downloaded from Gen Bank. A total of 233 species of 873 cytochrome C oxidases I gene (COI) sequence analysis of the structural characteristics of the gene. The results showed that there were 508 invariant loci, accounting for 82.1% of the total number of loci, 70 loci of transformation and 41 loci of transversions, accounting for 11.3% of the total number of loci % And 6.6% respectively. The intraspecific genetic distance was 0 ~ 0.019, the average genetic distance was 0.003 ~ 0.0034, the intraspecific genetic distance was 0.003 ~ 0.258, the average was 0.086 ~ 0.038. The phylogenetic tree shows that 227 species (97.4%) can be clustered into independent branches and have high support. On the basis of the DNA barcodes of 39 species of 11 genera in Brachycede family, 37 specific probes of 25 species were screened out. The success rate of this method for rapid identification of Brachyura species was 64.1% The results show that DNA barcode chip is used for the identification of Trichosanthes Branch has a certain feasibility.
