目的:建立肿瘤神经新生现象的体外研究模型,探讨在体外共培养模式下,人舌鳞状细胞癌细胞系HN-12对Sprague-Dawley(SD)大鼠三叉神经节(TG)突起生长的影响。方法:无菌条件下将HN-12细胞与SD大鼠TG接种于6孔板的Matrigel胶内进行共培养,TG单独培养作为对照组。分别于培养的第24、48、72、96、120 h拍照,测量实验组和对照组TG节神经突起生成的长度,利用SPSS19.0软件包检测2组神经突长度有无显著差异。结果:连续培养5 d发现,在共培养组,HN-12细胞对神经节突起的形成和生长具有明显的促进作用,与对照组相比有显著差异。并且,实验组中神经节突起具有明显的趋肿瘤细胞生长特性。结论:舌鳞癌神经新生现象体外培养模型成功建立。HN-12细胞具有促神经节突起生长的作用。
OBJECTIVE: To establish an in vitro model of neoplasia of tumor and to investigate the effect of human tongue squamous cell carcinoma cell line HN-12 on the growth of trigeminal ganglion (TG) in Sprague-Dawley (SD) rats in vitro co-culture model . Methods: HN-12 cells were inoculated into Matrigel of 6-well plates under aseptic conditions and were inoculated with TG in SD rats. TG alone was used as the control group. The 24th, 48th, 72th, 96th, 120th h were photographed respectively to measure the length of neurite outgrowth in experimental group and control group. SPSS19.0 software package was used to detect the difference of neurite length between the two groups. Results: After 5 days of continuous culture, HN-12 cells significantly promoted the formation and growth of ganglion neurons in co-culture group, which was significantly different from the control group. In addition, the ganglion neurites in the experimental group showed obvious tumor cell growth characteristics. Conclusion: The neuroblastoma model of tongue squamous cell carcinoma was successfully established in vitro. HN-12 cells have the role of promoting neurite outgrowth.
