目的:初步探讨β3GnT8/GalT7基因对HL60细胞体外侵袭的作用。方法通过脂质体介导将实验室构建好的β3GnT8真核正义表达载体pEGFP-C1-T8-Sense和反义表达载体pEGFP-C1-T8-AntiSense转染入HL60细胞;RT-PCR和Western blot检测浸润转移相关的功能基因TGFβ1、MMP2、MMP14在mRNA水平和蛋白质水平的表达变化;Transwell侵袭实验检测β3GnT8对HL60细胞侵袭转移能力的影响。结果随着β3GnT8表达上调,细胞侵袭能力高于对照组(P<0.05),MMP2、MMP14在mRNA和蛋白质水平的表达上调,而TGFβ1表达下调;随着β3GnT8表达下调,细胞侵袭能力下降,MMP2、MMP14在mRNA和蛋白质水平的表达下调,而TGFβ1表达上调。结论β3GnT8可在体外增强HL60细胞的侵袭能力,而该作用可能源于它能够诱导肿瘤侵袭相关蛋白的表达变化。“,”Objective To explore the effects of β3GnT8 gene on cell invasive potential of HL60 and its mechanism. Methods We transfected the recombinant plasmids pEGFP-C1-T8-Sense and pEGFP-C1-T8-AntiSense into HL60 with liposomes, and studied the variational characteristics of MMP2, MMP14 and TGF-β1 expression on mRNA and protein level with RT-PCR and Western blot. The invasive potential of the transfected cells was examined with the flow transwell plate. Results The expression ofβ3GnT8 increased in HL60 transfected with sense recombinant plasmid while the expression of MMP2 and MMP14 also upgraded.β3GnT8 expressed less in HL60 transfected with antisense recombinant plasmid while MMP2 and MMP14 also expressed weaker.β3GnT8 has negative correlation with TGF-β1.Cell invasion ability is higher than that of the control group (P<0.05). Conclusionβ3GnT8 can enhance HL60 cells in vitro invasion ability, and this effect may be derived from the expression changes that can induce tumor invasion related protein.
