在证明10ug/ml阿霉素诱导粘液表皮样癌MEC—1细胞凋亡的基础上,研究细胞凋亡过程中细胞周期变化,以利于临床用药。方法:通过显微镜观察细胞的形态变化和DNA电泳来观察图形,并依靠流式细胞术准确了解用药后细胞周期的变化。结果:光镜下见癌细胞凋亡改变,DNA电泳是典型梯状条图;流式细胞仪测得阿霉素作用5h时,细胞阻于G2期,24h时细胞阻于G1期。结论:阿霉素诱导MEC—1细胞凋亡初期,细胞阻于G2期,后期细胞阻于G1期。
In order to prove that 10ug/ml doxorubicin induces apoptosis of mucoepidermoid carcinoma MEC-1 cells, cell cycle changes during cell apoptosis were studied to facilitate clinical drug use. Methods: Observe the shape of the cells through microscope observation and DNA electrophoresis, and rely on flow cytometry to accurately understand the change of cell cycle after administration. RESULTS: Under light microscope, the apoptosis of cancer cells was observed. DNA electrophoresis was a typical ladder-like bar graph. Flow cytometry showed that when doxorubicin was used for 5 h, cell resistance was in G2 phase, and at 24 h, cell resistance was in G1 phase. Conclusion: Doxorubicin induces apoptosis of MEC-1 cells in the early stage, cell resistance in G2 stage, and late stage of cell resistance in G1 stage.
