肌腱膜纤维肉瘤癌基因同系物A(MafA)是特异表达于β细胞的一种转录因子,参与调节胰岛素合成、分泌和糖代谢等相关基因的表达。MafA特异结合于胰岛素基因C1元件,与结合于A1、E1元件的转录因子胰腺和十二指肠同源盒因子1、β细胞E盒反式作用因子共同调节胰岛素基因的转录。MafA在葡萄糖调节胰岛素基因表达过程中具重要作用,血糖变化可通过改变MafA的表达、稳定性及其与C1元件的结合活性调节胰岛素基因的转录。MafA的变异或缺失与糖尿病发生密切相关。
Apoptosis Fibrosarcoma Oncogene homolog A (MafA) is a transcription factor that is specifically expressed in beta cells and is involved in the regulation of insulin-related genes such as insulin synthesis, secretion and glucose metabolism. MafA specifically binds to the insulin gene C1 and interacts with the transcription factor pancreas and duodenal homeobox 1, β-cell E box transactivator that binds to A1 and E1 elements to regulate insulin gene transcription. MafA plays an important role in glucose regulation of insulin gene expression. Blood glucose regulation can regulate insulin gene transcription by changing the expression and stability of MafA and its binding activity with C1 element. MafA mutation or deletion is closely related to the occurrence of diabetes.
