目的探讨甘氨酸锌(glycine zinc)对甲型肝炎病毒(hapatitis A virus,HAV)抗原诱导小鼠体液免疫应答的影响。方法将ICR小鼠随机分为7组:甘氨酸锌(200μg,p H 6.5)+HAV组、甘氨酸锌(400μg,p H 6.5)+HAV组、甘氨酸锌(200μg,p H 7.5)+HAV组、甘氨酸锌(400μg,p H 7.5)+HAV组、生理盐水对照组(生理盐水200μl)、HAV抗原对照组(HAV抗原)和铝佐剂对照组(铝佐剂300μg+HAV抗原),每组8只。HAV抗原剂量均为18 EU,各组均经腹部皮下多点注射,200μl/只,免疫1次。免疫4、8、12、16周后,采用间接ELISA法检测小鼠血清中抗-HAV抗原特异性Ig G抗体水平,并进行安全性试验。结果除生理盐水对照组外,其他各组小鼠免疫4周后均产生抗-HAV Ig G,并随着时间的延长呈上升趋势,于免疫后第8周达峰值,此后逐渐下降。不同甘氨酸锌佐剂组的抗体水平均明显高于HAV抗原对照组。免疫后第8周时,铝佐剂对照组和甘氨酸锌佐剂组的抗体水平均显著高于HAV抗原对照组(P均<0.05);甘氨酸锌(400μg,p H 6.5)佐剂组抗体水平与铝佐剂对照组差异无统计学意义(P>0.05)。结论甘氨酸锌能显著增强HAV抗原诱导小鼠的体液免疫应答,其体液免疫增强效果与铝佐剂相当。
Objective To investigate the effect of glycine zinc on humoral immune response induced by hapatitis A virus (HAV) antigen in mice. Methods ICR mice were randomly divided into 7 groups: zinc glycine (200 μg, p H 6.5) + HAV group, zinc glycine (400 μg, p H 6.5) + HAV group, zinc glycinate (400 μg, p H 7.5) + HAV group, normal saline control group (saline 200 μl), HAV antigen control group (HAV antigen) and aluminum adjuvant control group (aluminum adjuvant 300 μg + HAV antigen) only. HAV antigen dose was 18 EU, each group were injected subcutaneously in the abdomen, 200μl / only, immunized once. After 4, 8, 12 and 16 weeks of immunization, the level of anti-HAV antigen-specific Ig G antibody in serum of mice was detected by indirect ELISA and the safety test was carried out. Results In addition to the saline control group, all the other groups of mice developed anti-HAV Ig G after 4 weeks of immunization and showed an increasing trend with time, reaching the peak at the 8th week after immunization, and then gradually decreased. Antibody levels in different glycine zinc adjuvant groups were significantly higher than the HAV antigen control group. Antibody levels in the aluminum adjuvant and glycine zinc adjuvant group were significantly higher than those in the HAV antigen control group (P <0.05) at week 8 after immunization. Antibody levels in the adjuvant group of zinc glycine (400 μg, p H 6.5) There was no significant difference between control group and aluminum adjuvant (P> 0.05). CONCLUSION: Zinc glycinate can significantly enhance the humoral immune response induced by HAV antigen in mice, and its humoral immune enhancement effect is comparable to that of aluminum adjuvant.
