目的 通过对同一来源的成猪胰岛采用两种不同的免疫隔离材料进行微囊化包裹 ,并植入糖尿病大鼠腹腔内加以比较 ,以明确两种方法间的优劣。方法 将同一来源的成猪胰岛分成三组 :未微囊化组、海藻酸钠微囊组和琼脂糖微囊组。 2 4只糖尿病大鼠随机分为 4组 :对照组、未微囊化胰岛移植组、海藻酸钠微囊化胰岛移植组和琼脂糖微囊化胰岛移植组 ,分别进行腹腔内胰岛移植。结果 4组糖尿病大鼠胰岛移植前血糖水平无显著差异 ,移植后 1周分别为 (2 2 6 7± 1 15)mmol/L ,(18 58± 2 6 2 )mmol/L ,(12 2 7± 1 36 )mmol/L和 (12 38± 2 6 8)mmol/L ,后 2组与前 2组相比均有显著差异 ,且生存期明显长于前 2组 ,但琼脂糖微囊化胰岛组的生存期明显短于海藻酸钠微囊化胰岛组 (分别为平均 57 10d和 145 17d)。结果 两种包膜材料制备的微囊化成猪胰岛均可存活于糖尿病大鼠体内并发挥生物功能 ,但海藻酸钠微囊化胰岛组较琼脂糖微囊化胰岛组的存活期明显延长。
OBJECTIVE To compare the advantages and disadvantages of the two methods by using microencapsulation of two different immunoisolated materials from the same origin of porcine islets and implanted into the abdominal cavity of diabetic rats. Methods The piglets from the same source were divided into three groups: non-microencapsulated group, sodium alginate micro-capsule group and agarose micro-capsule group. Twenty four diabetic rats were randomly divided into 4 groups: control group, non-microencapsulated islet transplantation group, microencapsulated alginate islet transplantation group and agar microencapsulated islet transplantation group, respectively, for intraperitoneal islet transplantation. Results There was no significant difference in blood glucose level before islet transplantation in 4 diabetic rats. After 1 week of transplantation, the blood glucose level was (2267 ± 1 15) mmol / L, (18 58 ± 2 6 2) mmol / L, (1227 ± 1 36) mmol / L and (12 38 ± 2 6 8) mmol / L, respectively. There were significant differences between the latter two groups and the former two groups, and the survival time was significantly longer than the first two groups. However, the agarose microencapsulated islets Group survival was significantly shorter than the alginate microencapsulated islets (mean 57 10d and 145 17d, respectively). Results Both microencapsulated porcine islets prepared from the two kinds of coated materials could survive in diabetic rats and exert their biological functions. However, the survival time of microencapsulated islets in the alginate microencapsulated islets was significantly longer than that of the microencapsulated islets.
