目的:探讨软肝冲剂治疗肝纤维化和肝硬化的机制是否与抑制TIMP-1的表达有关。方法:用CCl4诱导大鼠的肝纤维化模型(病理分期为0~Ⅴ期),将大鼠随机分成两组,分别用软肝冲剂和生理盐水治疗2个月后,对肝组织TIMP-1mRNA的表达进行分析,并检测羟脯氨酸(Hyp)的量,同时检测血清中TIMP-1和TGF-β1的浓度。结果:软肝冲剂能够降低大鼠肝组织中TIMP-1mRNA的表达和胶原蛋白的含量,同时血清中TIMP-1和TGF-β1较CCl4诱导的模型组和生理盐水治疗组显著降低(P<0·05)。结论:软肝冲剂治疗肝纤维化可能通过阻止TIMP-1的表达起作用。
Objective: To investigate whether the mechanism of Ruangan Granules in treating liver fibrosis and cirrhosis is related to inhibiting the expression of TIMP-1. METHODS: Rat models of hepatic fibrosis were induced with CCl4 (pathological stage 0 to V). Rats were randomly divided into two groups. After 2 months of treatment with Ruangan granules and physiological saline, TIMP-1 mRNA was detected in liver tissue. The expression was analyzed, and the amount of hydroxyproline (Hyp) was detected, and the concentrations of TIMP-1 and TGF-β1 in the serum were measured. RESULTS: Ruangan Granules could reduce the expression of TIMP-1 mRNA and collagen in rat liver tissue, and serum TIMP-1 and TGF-β1 were significantly lower than those in CCl4-induced model group and normal saline treatment group (P<0). ·05). Conclusion: Ruangan Granules may play a role in preventing hepatic fibrosis by blocking the expression of TIMP-1.
