以根癌农杆菌的Ti质粒为介导,将抗真菌病的几丁质酶基因,导入黑龙江省栽培大豆-东农37号,吉林28号等14个品种。从子叶节和下胚轴诱导出丛生芽,并再生植株,经PCR和DNA分子杂交检测,确认为转化植株。用花粉管通道导入法,直接导入几丁质酶基因,共获223粒种子,经PCR和DNA分子杂交检测的113株大豆幼苗中,有7株呈阳性反应,证明几丁质酶基因已导入并整合到大豆的基因组中。
Taking Ti plasmid of Agrobacterium tumefaciens as mediator, the antifungal chitinase gene was introduced into 14 cultivars of cultivated soybean Dongnong 37 and Jilin 28 in Heilongjiang Province. Clustered buds were induced from cotyledonary node and hypocotyls and regenerated plants were confirmed as transformed plants by PCR and DNA molecular hybridization. A total of 223 seeds were directly introduced into the chitinase gene by pollen tube channel induction. Among 113 soybean seedlings detected by PCR and DNA molecular hybridization, 7 showed positive reaction, indicating that the chitinase gene has been introduced And integrated into the genome of soybean.
