[目的]筛选胃癌患者血清肿瘤标志物,用于胃癌早期诊断。[方法]应用表面增强激光解析离子化飞行时间质谱技术(Surface Enhanced Laser Desorption/Ionization Time-of-flight Mass Spectrometry,SELDI-TOF-MS),检测32例胃腺癌、22例消化性溃疡、32例胃炎患者和30名正常人(对照)的外周血清标本,筛选胃癌相关特异蛋白峰。[结果]与正常对照相比较,发现质荷比为2953、3267、5341、5912、5927的蛋白质分子在胃癌患者血清中上调,质荷比为4059、4213、4270、7160的蛋白质在胃癌患者中下调。4059、5341、5912、3267、4059及5341的蛋白质组成的波谱模型可以准确鉴别胃癌与正常对照。其中质荷比为5912的蛋白质在胃癌患者血清中有很高的表达,敏感性为81.25%,特异性为56.67%。4059的波谱峰可以在正常组中显示,但在胃癌组中几乎消失。[结论]SELDI-TOF-MS技术是一种快速、简便、易行且高通量的分析方法。本研究筛选出的4059、5341、5912、3267、4059及5341低分子量蛋白质有可能成为潜在的诊断胃癌的血清生物标志物。
[Objective] To screen the serum tumor markers of gastric cancer patients for the early diagnosis of gastric cancer. [Method] The surface enhanced laser desorption / ionization time of flight mass spectrometry (SELDI-TOF-MS) was used to detect 32 cases of gastric adenocarcinoma, 22 cases of peptic ulcer and 32 cases of gastric ulcer Gastritis patients and 30 normal people (control) of peripheral blood serum samples, screening for gastric cancer-specific protein peaks. [Results] Compared with the normal control, protein molecules with mass-to-charge ratios of 2953, 3267, 5341, 5912 and 5927 were found to be upregulated in the serum of gastric cancer patients. The proteins with mass-to-charge ratios of 4059, 4213, 4270 and 7160 were found in gastric cancer patients Down. 4059, 5341, 5912, 3267, 4059 and 5341 protein profiles can accurately identify gastric cancer and normal controls. The protein with a mass-to-charge ratio of 5912 was highly expressed in the serum of patients with gastric cancer, with a sensitivity of 81.25% and a specificity of 56.67%. The peak of 4059 can be displayed in the normal group but almost disappears in the gastric cancer group. [Conclusion] The SELDI-TOF-MS technique is a fast, simple, easy and high throughput analytical method. The 4059, 5341, 5912, 3267, 4059 and 5341 low molecular weight proteins screened in this study may become potential biomarkers for the diagnosis of gastric cancer.
