目的:观察加味逍遥散对抑郁大鼠行为学及海马组织炎症因子表达的影响,探讨其可能的作用机制。方法:将60只SD大鼠随机分为空白组、模型组、氟西汀组、加味逍遥散低、中、高剂量组,每组10只。除空白组外,其余各组均腹腔注射LPS建立抑郁大鼠模型。氟西汀组每天灌胃盐酸氟西汀1.54 mg/(kg·d),加味逍遥散低、中、高各剂量组每天分别灌胃加味逍遥散2.5 g/(kg·d)、5 g/(kg·d)、10 g/(kg·d),空白组及模型组灌胃等体积生理盐水。通过旷场实验及强迫游泳实验观察大鼠行为学的变化,采用高效液相-质谱联用(HPLC-MS/MS)技术检测海马组织中5-羟色胺(5-HT)含量,实时荧光定量PCR(RT-PCR)检测海马组织IL-1βm RNA、IL-6 m RNA、TNF-αm RNA表达水平。结果:与空白组比较,模型组大鼠的起立数、穿格数、修饰数均明显减少,强迫游泳不动时间延长,海马组织5-HT浓度显著降低,而IL-1βm RNA、IL-6 m RNA、TNF-αm RNA表达显著升高,差异均有统计学意义(P<0.01)。与模型组比较,氟西汀组、加味逍遥散高剂量组大鼠起立数、穿格数、修饰数显著增加,强迫游泳不动时间缩短,海马组织5-HT浓度显著升高,而IL-1βm RNA、IL-6 m RNA、TNF-αm RNA表达量明显降低,差异均有统计学意义(P<0.05,P<0.01)。结论:加味逍遥散对LPS诱导抑郁模型大鼠具有明显抗抑郁作用,通过抑制IL-1βm RNA、IL-6m RNA、TNF-αm RNA的过度表达,增加大鼠海马组织5-HT的含量来改善大鼠抑郁症状。
Objective: To observe the effects of Jiawei Xiaoyaosan on the behavior and the expression of inflammatory cytokines in hippocampus of depressed rats, and to explore its possible mechanism. Methods: Sixty SD rats were randomly divided into blank group, model group, fluoxetine group, plus Xiaoyaosan low, medium and high dose group, 10 rats in each group. Except the blank group, all the other groups were given intraperitoneal injection of LPS to establish a rat model of depression. In fluoxetine group, fluoxetine hydrochloride 1.54 mg / (kg · d) was given daily, while Jiawei Xiaoyaosuan low, medium and high dosage groups were fed with 2.5g / (kg · d) (kg · d) and 10 g / (kg · d) respectively. The rats in the blank group and model group were given gavage with equal volume of normal saline. The behavioral changes of rats were observed by open field test and forced swimming test. The content of 5-HT in rat hippocampus was detected by HPLC-MS / MS. Real-time fluorescence quantitative PCR The mRNA expression of IL-1βmRNA, IL-6mRNA and TNF-αmRNA in the hippocampus were detected by RT-PCR. Results: Compared with the blank group, the upright number, the number of passing through and the number of modification of the model group were significantly decreased, forced swimming time was prolonged, the 5-HT concentration in hippocampus was significantly decreased, while IL-1βmRNA, IL-6 The mRNA expression of m RNA and TNF-αm were significantly increased, the difference was statistically significant (P <0.01). Compared with the model group, fluoxetine group and Jiawei Xiaoyaosan high-dose group increased number of standing, passing number and modifying number, forced swimming time was shortened, 5-HT concentration in hippocampus increased significantly, while IL- 1βmRNA, IL-6mRNA and TNF-αmRNA were significantly decreased, the differences were statistically significant (P <0.05, P <0.01). CONCLUSION: Jiawei Xiaoyaosan has obvious anti-depressant effect on rats with LPS-induced depression and can be improved by inhibiting the over-expression of IL-1βmRNA, IL-6mRNA and TNF-αmRNA and increasing the content of 5-HT in hippocampus of rats Depression symptoms in rats.
